Technology for creating transgenic laboratory animals
Summary
The technology is aimed at creation of transgenic laboratory animals using microinjection into pronuclei of fertilized zygotes of mouse gene structures in the form of expression cassettes containing sections of human genes under regulation of promoters providing for the tissue-specific expression of the target gene. The technology includes stages of creating transgenic mice via delivering expression vectors to the genome of animals, created based on plasmid DNA and carrying sequences of human genes under control of non-viral regulatory elements of various origins: creation and production of genetic and engineered structures aimed at obtaining transgenic mice; preparative isolation and purification of DNA preparations containing gene structures; preparation of experimental animals for introduction of exogenous DNA; work on delivery of gene structures into the genome of mice using method of pronuclear microinjection.
Technology Benefits
The economic effect of created gene structures will be about 250 thousand US dollars per 1 gene structure.[br][/br]Obtaining transgenic producing animals with pharmacologically active human protein may lay the foundation for development of domestic pharmaceutical substances of recombinant human proteins and biologic preparations of new generation, contributing to import substitution and cost reduction by 0.5–2 times of some antimicrobial and thrombolytic preparations.
